These compounds were potent cytotoxic agents with IC50 values in the low micromolar concentration range and demonstrated enhanced selectivity in receptor-negative cells compared to SAPs and 4-thiazolidinone amides. During the course of structure-activity relationship studies of above class of compounds we were interested to investigate the effect of heteroatoms in the lipophilic alkyl side chain on potency and selectivity. It is also well known that polyamine containing compounds exhibit a number of biological activities and have been utilized as chemotherapeutic agents. Due to these reasons, we designed and prepared a series of compounds containing serine, 4-thiazolidinone and thiazolidine-4-carboxylic acid as head groups conjugated with naturally occurring polyamines like putrescine, spermidine and spermine. Short chain polyamines conjugated to serine, thiazoldinone, and thiazolidine carboxylic acid did not show activity up to 100 M. As the length of polyamine moiety increased, cytotoxicity also increased in prostate cancer cells.
